The role of necroptosis in disease and treatment

Necroptosis is activated by a unique caspase-independent signaling pathway and mainly relies on RIPK1/RIPK3/MLKL complex. Over the past decade, necroptosis has been found widely involved in life process of human beings and animals. In this article, we attempt to explore therapeutic prospects of the necroptosis regulators in health maintenance and disease treatment by describing the molecular mechanism of necroptosis and the role it played in disease and tissue homeostasis, summarizing the research and clinical applications of corresponding inhibitors and inducers which include small molecule inhibitors, chemicals, Chinese herbal extracts, and biological agents in the treatment of a variety of diseases including cancer, inflammation, infection, ischemia-reperfusion injury, neurodegenerative diseases in various organs including cardiovascular, pulmonary, liver, pancreatic, renal, enteric, ocular, joint, and skin.
NF-κB signaling in inflammation and cancer

This year (2021) marks the 35th anniversary of the discovery of NF-κB. With so many years of in-depth research on NF-κB, people have realized that the NF-κB signaling pathway plays an important role in inflammation, immunity, cell survival and proliferation. This review summarizes the relevant knowledge of the NF-κB signaling pathway, inflammation, and cancer. The progression of the translational study and drug development targeting NF-κB for inflammatory diseases and cancer treatment and the potential obstacles will also be discussed.

Autophagic activation of IRF-1 aggravates hepatic ischemia–reperfusion injury via JNK signaling

The expression of interferon regulatory factor-1 (IRF-1) is related to the high autophagy activity and aggravates the liver damage, which is offset by IRF-1 depletion in vivo or in vitro. IRF-1 can activate autophagy to aggravate hepatic ischemia–reperfusion via regulating JNK pathway.
Lomerizine 2HCl inhibits cell proliferation and induces protective autophagy in colorectal cancer via the PI3K/Akt/mTOR signaling pathway

Lomerizine 2HCl, which is an approved prophylactic drug for migraines, was selected for our studies. In vitro and in vivo functional assays were performed, and the data suggest that lomerizine 2HCl inhibits cell growth and promotes apoptosis in CRC cells. Moreover, lomerizine 2HCl inhibits cell migration and invasion of CRC. RNA sequencing analysis and Western blotting confirmed that lomerizine 2HCl can inhibit cell growth, migration, and invasion via the PI3K/AKT/mTOR signaling pathway and induces protective autophagy in CRC. Meanwhile, autophagy inhibition by 3-methyladenine (3-MA) increases lomerizine 2HCl-induced cell apoptosis. Taken together, these results suggest that lomerizine 2HCl is a potential therapeutic agent, and the combination of lomerizine 2HCl and autophagy inhibitors may serve as a novel strategy to increase the anticancer efficacy of agents in the treatment of CRC.
Lycopene antagonizes lead toxicity by reducing mitochondrial oxidative damage and mitochondria-mediated apoptosis in cultured hippocampal neurons

In this study, we demonstrated the neuroprotective effects of lycopene in primary cultured rat hippocampal neurons challenged with lead. The protective effects of lycopene may be attributable to its roles in reducing mitochondrial oxidative damage and mitochondria-mediated apoptosis.

Neutrophils help cancer cells metastasis in distant organs. Clinical multidrug-resistant (MDR) S. aureus infection enhances cancer recurrence via orchestrating the NETosis network to promote metastasis. Targeting autophagy and/or NETs in bacterial infection may be useful for patient prognosis and may reduce metastasis.
Tyrosine phosphatase SHP2 exacerbates psoriasis-like skin inflammation in mice via ERK5-dependent NETosis

Single-cell RNA sequencing and experimental verification were combined to announce that SHP2 aggravates psoriasis-like skin inflammation in mice via ERK5-dependent NETosis. During this process, inflammatory cytokines: TNF-α, IL-1β, IL-6, IL-17a, and CXCL-15 were infiltrated in skin and contributed to psoriasis. Our study provides evidence for the role of SHP2 in NETosis and psoriasis. SHP2 may be a potential therapeutic target for the treatment of psoriasis.
Clinical benefit of neoadjuvant anti-PD-1/PD-L1 utilization among different tumors

Twenty studies were included in this meta-analysis. The pooled CRR of triple-negative breast cancer and urothelial carcinoma was 0.569 (95% CI 0.514, 0.624) and 0.320 (95% CI 0.234, 0.407), respectively. The pooled MRR of lung cancer was 0.471 (95% CI 0.367, 0.575). In conclusion, neoadjuvant anti-PD-1/PD-L1 therapy received satisfactory clinical results in these tumors.

Engagement of CAR-T cells through PD-1/PD-L1 ligation. We speculate that PD-L1-expressed CAR- T cells lead to a suppressive signal by PD-1/PD-L1 engagement. From there, T cell-associated PD-L1 is a major therapeutic target of immune checkpoint blockade.
